Scientists help identify a missing link in taste perception

Working with a multidisciplinary consortium of 19 researchers from nine institutions, Monell scientists have provided critical information to identify CALHM1, a channel in the walls of taste receptor cells, as a necessary component in the process of sweet, bitter, and umami (savory) taste perception.

When sweet, bitter and umami molecules reach the tongue, they activate taste receptors in specialized cells called Type II taste cells. "The question that the consortium wanted to answer is, 'how do these taste cells tell the brain that they have detected something?' said Monell taste biologist Michael G. Tordoff, PhD. "This question has been a longstanding missing link in our understanding of taste perception."

The scientists already knew that activation of taste receptors on Type II cells initiates a complex chain of events inside the taste cells. What they found, as reported in the current issue of Nature, is that the final step involves the opening of a pore formed by CALHM1 in the taste cell membrane. The open channel allows molecules of the neurotransmitter ATP to leave the taste cell and relay a signal to adjacent nerve cells connected to the brain.

Monell molecular neurobiologist Ichiro Matsumato, PhD, contributed to the work by showing that the gene for CALHM1 is expressed in Type II taste cells, but not in other types of taste tissue. "Our findings demonstrate that the CALHM1 pore is localized specifically in cells that detect sweet, bitter and umami taste," said Matsumato.

The necessity of CALHM1 for the ability to taste sweet, bitter, and umami was demonstrated in behavioral tests performed by Tordoff. Reasoning that mice lacking the CALMH1 channel would not be able to release ATP to send information about sweet, bitter and umami taste detection to the brain, Tordoff tested the taste preferences of Calhm1 'knockout' mice. Engineered by co-author Philippe Marambaud, PhD, of the Feinstein Institute for Medical Research, the knockout mice lack the gene that codes for CALHM1.

"Like humans, mice with an intact CALHM1 gene avidly drink sucrose and other sweeteners, and avoid bitter compounds such as quinine. However, mice lacking CALHM1 are very unusual," said Tordoff. "These mice treat sweeteners and bitter compounds as if they were water. They behave as if they can't taste them at all."

Responses to salty and sour tastes were not affected by the missing gene because perception of these taste qualities is mediated via a different set of taste cells.

In combination with electrophysiological data contributed by collaborators from other institutions, the findings demonstrate that pannexins and connexins, channel proteins previously thought to be involved in ATP release from taste cells, actually are not necessary for this to happen. "This paper provides compelling data to overturn the previous hypothesis," noted Matsumato. "It's part of what makes science so exciting."

Kevin Foskett, PhD, professor of Physiology at the Perelman School of Medicine, University of Pennsylvania, who with Marambaud is a senior author on the paper, recently identified CALHM1 as an ATP channel and speculated that that it might be involved in taste. "This is an example of a bona fide ATP ion channel with a clear physiological function," said Foskett. "Now we can connect the molecular dots of sweet and other tastes to the brain."

In addition to revealing CALHM1's critical role in sweet, bitter, and umami taste perception, the current work also lends insight into the channel's overall function in other tissues. Originally thought to control calcium levels inside cells, CALHM1 (calcium-homeostasis-modulator-1) may also contribute to ATP-mediated intercellular communication in the brain and elsewhere.

E-hail app to order yellow cabs in NYC blocked by judge

NEW YORK (Reuters) - A pilot program that would allow people in New York City to hail yellow cabs using a smartphone app was blocked by a judge on Thursday, handing a victory to private car companies that have said the program threatens their business.

Manhattan Supreme Court Justice Carol Huff granted a request from the livery companies to freeze the new "e-hail" program that had been scheduled to begin as early as Friday.

The order will be in effect until at least March 18, when the two sides are due to return to court to argue the merits of a lawsuit brought by the livery companies against the city.

The pilot program would allow people to use a phone application to request a yellow cab ride. Taxi drivers would be able to receive such requests and confirm pickup locations.

Private car companies rely on prearranged pickups. Under New York's two-prong cab system, yellow cabs handle street hails but cannot accept phone pickups; livery cars do the opposite.

The program would be an option for New Yorkers accustomed to standing in the street and flagging down the city's approximately 13,000 yellow cabs. E-hail apps like Uber and Hailo have become popular in other cities.

The maternal effect: How mother deer protect their future kings

Mar. 5, 2013 ? Do mothers invest more care in their sons if they believe their child is destined to be a king, president or a high-powered leader? The answer is definitively yes -- as long as those mothers and their sons happen to have hooves.

New research from BYU reports that, just like the classic tale of Bambi, females from the deer family are more likely to invest more in the survival and health of their male offspring if there is a good chance those sons will become a "Great Prince of the Forest."

"Our research demonstrates clearly that a mother's investment in her offspring was evident during adulthood, even though offspring live independently of their mothers from a very young age," said Brock McMillan, associate professor of wildlife ecology at BYU.

The comprehensive study of deer and elk from the Intermountain West found that the most dominant males at the time of death were those who were born into the most favorable maternal conditions 5 to 15 years before.

While favorable maternal conditions are largely tied to the health of the expectant mother, there are additional elements at play.

A mother's investment happens both in the womb and during the first few months of life. During those early stages of life, mothers take care to provide more excellent nourishment through lactation as well as better habitats for the baby.

"Male deer and elk live independently of their mothers for several years in highly variable environments," McMillan said. "They live through severe winters with deep snow and little to eat, dry summers with poor quality food and years of injuries and ailments associated with everyday life. Even with many years of exposure to the environment, the maternal effect was still evident."

The study, appearing in the current issue of scientific journal PLOS ONE, provides new information for how deer and elk populations propagate.

Whereas deforestation, human-encroachment into natural habitats, increased predators, and several other factors have been thought to play the most prominent roles in determining deer and elk population trends, this study implies that an equally important factor is the health and nurturing of the mothers years before.

"For those unconcerned about deer or elk, this research can be a reminder that similar research exists for humans," said student Eric Freeman, lead author on the study. "Conditions experienced in utero affect offspring throughout their life even in long-lived species."

With the study, Freeman joined the throngs of BYU students before him who completed peer-review research as undergraduates.

"In many ways, this research was the most important part of my undergraduate education," Freeman said. "I became a much better thinker and writer as a direct result of my work on this research with my advisors."

Freeman is currently a graduate student in the College of Life Sciences studying mule deer fawns. He plans to pursue a PhD with the goal of eventually teaching and conducting research at a university.

Wildlife ecology professor Randy Larsen is also a co-author on the study. McMillan and Larsen are professors in the department of Plant and Wildlife Sciences.

Detroit, Host Of The Olympics In 2024? No Thanks, Says Mayor Dave Bing

Grappling with a financial emergency, Detroit is waiting to hear the state's plans for the city's future. But there's one plan that definitely isn't in the cards: hosting the 2024 Olympic Games.

Last month, the United States Olympic Committee sent letters to the mayors of 35 cities to see if they were interested in pursuing an Olympics bid. Those chosen included the nation's 25 largest cities, of which Detroit is one. In a statement, Mayor Dave Bing responded to the USOC query with a polite but resounding no, citing the large costs involved.

"With our rich history of hosting major events and Detroit?s prime location on an international border, we recognize that this makes Detroit an appealing candidate to potentially host the 2024 Olympics and Paralympics," he said. "Unfortunately, due to the timing and uncertainty of Detroit?s long-term financial stability, we must respectfully decline to participate.?

Surely some shot put, gymnastics and swimming fans will be disappointed in Bing's choice -- and particularly one group that has been using Facebook to rally support for an Olympics bid since last summer.

But the requirements for host cities, including a $10 million upfront bid processing payment, $3 billion operating budget commitment, construction of an Olympic Village for 16,500 athletes and, perhaps the biggest hurdle, a workable transportation system, make it hard to imagine Detroit being in shape to host the Olympics in a mere 11 years.

It's not clear that USOC had real hopes for Detroit's potential. According to The New York Times, most of the wide range of cities that were selected are likely already "virtually disqualified." Last summer, USOC past president Bill Martin told WXYZ it would be a "huge leap" for Detroit to host the Olympics.

At least the city had the opportunity to be the snubber, for once. In the past, Detroit's bids to host the Olympics have been rejected seven times.

There is one small chance Detroit hasn't heard the last of this Olympics talk. The group that has attempted to rally support for the 2024 games considered pitching the plan to the individual selected to be Detroit's emergency financial manager. And they have their fingers crossed for another long shot -- that Gov. Rick Snyder picks former presidential candidate Mitt Romney to run the show in Detroit. After all, he ran the 2002 Winter Olympics in Salt Lake City.

Romney in charge of Detroit (where he won 2 percent of the vote in November) and planning the city's Olympics? That's even less likely than these tongue-in-cheek suggestions for locations around town for Olympic events.
